Description of the projectPlease, provide project background information for the interested students:

Plants, insects, animals and humans produce natural peptide antibiotics in their secretions. In theory, producing these peptide antibiotics with Recombinant DNA Technology is possible.

What is the aim of the project?

The aim of the project is determine motifs of peptide antibiotics and produce them with recombinant DNA technology.

There will be lectures and practical training about the basis of recombinant DNA technology and peptide antibiotics

What is the role of the student and what is expected from him/her during the research exchange?

Learn and apply microbiology and molecular biology techniques like culturing bacteria, performing susceptibility tests, plasmid DNA isolation, pouring and running electrophoresis gels, DNA amplification by PCR.

 

What should be the outcome of the student’s participation on the research exchange project (paper, poster etc)?

The student will get theoretical and practical training on microbiological and molecular biological research techniques. The results of the research are expected to be presented as a poster in a scientific meeting.

What are the practical skills and the knowledge the student will acquire during the exchange program?

Microbiology and molecular biology techniques like culturing bacteria, performing susceptibility tests, plasmid DNA isolation, pouring and running electrophoresis gels, DNA amplification by PCR.

The students are expected to have basic knowledge in biology and chemistry. There are no special legal requirements.

D. fragilis is an intestinal parasite with known pathogenic effects. However, its conventional diagnosis using examination of stained stool smeas with trichrome, Giemsa reveals unsatisfactory results and underestimates its prevalence. Real time PCR is a highly sensitive and specific method and has been applied maybe to all microorganisms_Using this method, the prevalence of D. fragilis is intended to be unveiled in public and inchildren as well________________________________________________________________________

Carbapenemases are enzymes which break down carbapenem group of antimicrobials. Carbapenems have been widely used globally in the treatment of ESBL (extended-spectrum beta-lactamase) producing Enterobacteriaceae. However carbapenemases have occured and the spread of carbapenemase-producing Enterobacteriaceae isolates became a global problem. Various studies from Turkey have identified OXA-48 and IMP as the dominant carbapenemase-encoding genes in Turkey.

 

What is the aim of the project?

At the Acibadem Labmed Clinical Laboratories which receives clinical specimens from various cities in Turkey, it’s aimed to investigate the mechanisms responsible for carbapenem resistance in Escherichia coli and Klebsiella pneumoniae.

 

What techniques and methods will be used?

General culture techniques for bacteria, antimicrobial susceptibility testing (automated device, disk diffusion, broth microdilution), DNA extraction from bacteria, polymerase chain reaction (PCR).

Yes, the student will be provided with relevant literature and the materials and methods used in the study will be explained to the student prior the lab work during sessions.

 

What is the role of the student and what is expected from him/her during the research exchange? (minimum 3 lines)

Performing necessary procedures (inoculation of plates, performance of antimicrobial susceptibility tests, DNA extraction from colonies grown on Petri plates, preparation of PCR mixture, agar gels and evaluation of results). Entering the data on Excel file.

 

What should be the outcome of the student’s participation on the research exchange project (paper, poster etc)?

Experience in lab activities to investigate highly resistant bacteria, also depending on the results of the study, an international poster presentation can be anticipated.

 

What are the practical skills and the knowledge the student will acquire during the exchange program?

Basic rules to work in a bacteriology laboratory, aseptic techniques, subculturing bacteria, use of materials and devices such as inoculating loop, cotton swab, nephelometer, thermal cycler, electrophoresis tank.
